چکیده

Abstract Pentatricopeptide repeat (PPR) proteins are helical repeat-proteins that bind RNA in a modular fashion with sequence-specificity can be manipulated by the use of an amino acid code. As such, PPR repeats promising scaffolds for design binding synthetic biology applications. However, vivo functional capabilities artificial built from consensus motifs just starting to explored. Here, we report functions protein, dPPRrbcL, made were designed sequence near 5? end rbcL transcripts Arabidopsis chloroplasts. We used complementation assay demonstrate this protein bound its intended target specificity and it substituted natural stabilizing processed mRNA. targeted second analogous petL UTR, where native PGR3, albeit inefficiently. These results showed engineered functionally mimic class serve as physical barriers against exoribonucleases.

Chloroplast RNA editing by pentatricopeptide repeat proteins.

Pentatricopeptide repeat (PPR) proteins function in multiple processes in mRNA maturation in plastids and mitochondria (reviewed in Schmitz-Linneweber and Small, 2008). RNA maturation in chloroplasts is complex, and PPR proteins act in cisand trans-splicing, translation, RNA cleavage, RNA stabilization, and RNA editing. This large protein family, with 450 members in Arabidopsis, is characterize...

Chloroplast RNA-binding and pentatricopeptide repeat proteins.

Chloroplast gene expression is mainly regulated at the post-transcriptional level by numerous nuclear-encoded RNA-binding protein factors. In the present study, we focus on two RNA-binding proteins: cpRNP (chloroplast ribonucleoprotein) and PPR (pentatricopeptide repeat) protein. These are suggested to be major contributors to chloroplast RNA metabolism. Tobacco cpRNPs are composed of five diff...

Pentatricopeptide repeat proteins

Mitochondria and chloroplasts host genomes inherited from their bacterial ancestors. Their biogenesis thus requires fully functional gene expression machineries. RNA metabolism in organ-elles is particular and has attracted considerable attention since it combines prokaryotic features with unique traits that evolved in their eukaryotic host cell. For a long time, most of the molecular factors g...

Conserved domain structure of pentatricopeptide repeat proteins involved in chloroplast RNA editing.

The pentatricopeptide repeat (PPR) proteins form one of the largest families in higher plants and are believed to be involved in the posttranscriptional processes of gene expression in plant organelles. It has been shown by using a genetic approach focusing on NAD(P)H dehydrogenase (NDH) activity that a PPR protein CRR4 is essential for a specific RNA editing event in chloroplasts. Here, we dis...

HCF152, an Arabidopsis RNA binding pentatricopeptide repeat protein involved in the processing of chloroplast psbB-psbT-psbH-petB-petD RNAs.

The psbB-psbT-psbH-petB-petD operon of higher plant chloroplasts is a heterogeneously composed transcriptional unit that undergoes complex RNA processing events until the mature oligocistronic RNAs are formed. To identify the nucleus-encoded factors required for the processing and expression of psbB-psbT-psbH-petB-petD transcripts, we performed mutational analysis using Arabidopsis. The allelic...
